Clean the lens (only with lens cleaning solution!)
Then take another picture of the sky.
If it's gone, then it was dust or something.
Otherwise, there may be a worse problem.

given the focus, it could be a small hair on the CCD... you can activate the mirror lock function and blow a bit of *clean* air in it, then check if that fixed it.
Next time you swap lenses, try to keep dirt away...

No, it all depends on the light conditions and focus. If you say you only saw it in that pic, that's even more reason to suspect dust on the CCD (or maybe lens, inside side)
My camera currently also has dirt on the CCD, but as it only shows up in certain pics, I'm too lazy to have it cleaned
